Your MluI site within the 5' end of the ORF sounds like a DAS (Downstream
Activation Site). Have you characterized this element yet? Does it work?
We characterized the LPD1 gene and showed that there are both activation
and repression sites within the ORF (Sinclair et al,MCB, Jan 1994). When I
presented this work in 1992 at the Vienna Conference many people were sceptical
but an equal number of people also said that they have sites within their ORF.
I would be very interested if any more researchers have found the same
phenomena.
