We are trying to use the one hybrid system to clone a particular
DNA-binding factor from mammalian c-DNA/GAL4 activation domain libraries.
Does anyone have a gal4, gal80, leu2, trp1, ura3, his3 strain that we could
use for this?
Does anyone have an ARS/CEN expression vector that is URA3 containing a
constitutive promoter (eg. ADC1) that we could use. An ideal vector would
contain unique HinDIII and EcoRI sites downstream of the promoter and a
termination sequence?
We need such a vector to express an already cloned cDNA that is needed for
DNA-binding by the product of the cDNA we are trying to clone. The marker
has to be URA3 since we are using all the other markers in the strain.
We would very much appreciate any help we can get with either of these
