1) Yeah, even 150 basepairs on either end is ok (e.g. the PvuII sites adjacent to the polylinker in blue white vectors). 2) Yeah, we would be very interested as well if there is some concensus on how well the PCR trick works in general (under conditions where you don't know the KO phenotype ahead of time).
