Dear buddies,

I suspect that the reason why minimal plates won't sometimes set properly
is indeed hydrolysis of the agar. It makes a difference when you autoclave
takes too long to get to the right temperature - (sorry: I meant YOUR auto-
clave but I cannot return to correct it). But I also noticed that you
boil the sugar together with the YNB; so I suspect that your plates are
occassionaly not only mushy but also dark. It is always better to sterilize
the carbon source separately and mix it to the rest immediately before
pouring the plates (even for YEPD). Not only the plates look better this
way, but also the yeasties grow MUCH better on them!
