cloning a CEN-linked gene

Dear David,

sorry - I can't provide you with a CEN-marked yeast strain, but I wonder
whether it wouldn't be worth trying to clone your CEN-linked gene out of
a 2 micron library. Albeit there is a risk of picking overexpression
suppressors, I think that quite a lot of genes have been cloned this
way in the past. Besides, if the reason for your cloning problems indeed
is the close linkage to CEN, the resulting suppressor plasmid will anyway
be an YCp...

Another possibility, of course, is that someone or sometwo or somemore
out there in the net collected CEN-spanning cosmids for all the yeast
(sorry: BUDDING yeast) chromosomes - perhaps during the sequencing
project.

Good luck! Fatima
