Hello Everyone!
We have been doing b-gal filter assays for two hybrids systems but we find
these assays to be inconsistent. Sometimes they work beautifully and other
times we see no color development, and this is when using the same
transformants. Does anyone know what factors may influence these assays such
as growth period, media components, filter type, b-gal concentrations, etc. If
anyone is doing b-gal filter assays that consistently work, please share your
secrets.
We are also interested in trying to grow our yeast on b-gal plates, but we have
heard that the results on plates can be more inconsistent than filter assays.
Does anyone have any experince with b-gal plates?
