HEllO to all
jbhogen at merle.acns.nwu.edu wants to know about Yeast Electroporation!
Check out Manivasakam and Schiestl 1993 NAR 21:4414-4415
This is a procedure that the authors say can give up to 10^6 TRAFOs/µg.
We have used it and found that EPing yeast is highly variable. Each strain
is different! Some do well, like GGY1::171 (Stans Fields two hybrid
strain), and others just the shits.
Another Very good paper With numerous TRAFO methods is
Schiestl, Manivasakam, Woods and GIETZ, 1993 Introducing DNA into
yeast by transformation METHODS:A companion to Methods in Enzymology 5:
79-85.
There are both EP and LiAc/SS-DNA/PEG methods that give up to 5 x 10^6
TRAFO/µg of input DNA. ALSO see references within!
GOOD Luck with your TRAFOs
