Dear John et al.,
might be it sounds disappointing, but - at least according to my
experience - the only really good and almost 100% reproducible
protocol giving high efficiency is the good old tedious spheroplast
method (followed by lithium acetate transformation - I never got
electroporation working reliably). If you are interested, I'll
send you the protocols.
