Indeed, there is no method for absolute protein determination that could
be performed at the sample size required in a modern enzymological lab.
Absolute means that you do not need reference standards. Also, many
commonly used substances (detergents, reducing agens, complex formers,
urea) interfere with many protein assays.
So in a word: the situation is hopeless, but not serious.
