This is koteswara rao , I am facing the problem of
coupling reaction as well as identification of the hapten + protien
coupling.
Basically I am using a chemical molecule ( molecular weigth of 261 ) to
raise the antibodies. The molecule contains amino group, now I wanted
to couple the molecule with a carrier molecule (BSA / KLH ) by
conjugation. I came to know there are some protocols available to
couple with glutaraldehyde and EDC for combine the amino as well as
carboxyl groups respectively. It seems there are some dis advantages by
these methods. So I wanted to know the protocol with advantages and
drawbacks. My second question is how to detect the coupled molecule
from the mixture like how much amount has been coupled ? can any one
help me out to get some simple protocols?
