Hi,
I am trying to use hydroxylamine cleavage to cut a tag off a small
protein. Within 3 hours of starting the cleavage rxn my protein ppts
(final conc, 2M HA, 4.8M guanidine, 20mM Ches). The ppt seems to be
occuring before cleavage takes place. I have tried adding different
concentrations of DTT and changing the pH of the reaction ( I usually
use pH9.5) but have not been able to inhibit the ppt of the protein.
Any suggestions?
