I assume you are adding IPTG to your medium. The pET vector series
has a T7 promoter, which will normally not be recognized by E. coli
polymerases. Only when the T7 polymerase is expressed will your gene
be expressed. The T7 polymerase is under control of a Lac promoter,
which is positively regulated by the addition of IPTG.
