mitochondrial respiratory chain complexis quantification

Hello to all, 
I'm just new about proteomics and I'd like to quantify the subunits of mt complexes codified by mtDNA. Is there somebody who have experience in this?
I'm going to isolate mitochondria from platelets and use 2D-electrophoresis then mass analysis. 
Do you know how many cells (or mitochondria) I need to have good results?
Do you suggest other methods?
