MBP-cleavage site-protein of interest-Hisx6 sandwich is very good. We use
TEV protease cleavage site for obvious reasons. This sandwich is trivial to
construct. As to expression strain - usually BL21 DE3 with RIL or Rosetta
rare codon booster works well.
