Sephacryl 500 column problem...

Your column is probably losing bead shape due to rapid changes in solvent.
Multiple transitions from organics into water will do that to a brittle
resin, and Seph 500 is definitely brittle. If you have clogging, the
situation will rapidly become worse. Keep in mind that some of your samples
may have aggregates that remain forever bound to the resin.
