Have you tried a prediction service for looking for PTM sites? Some are
not that easy to find by hand.
If you are suspecting glycosylation, you could try to remove it
enzymatically. What about degradation ? Are the proteins produced
recombinantly?
Could you post the sequence surrounding the AA difference?
