what is the composition of the sample buffer (sophie) ?
I think U try to get a native protein by using triton in your extraction
(membrane proteins ?)but I was trying this detergent and it can trouble the
SDS-PAGE at high concentration (cmc ~0.025% for TX100).
like patrick, U can use Bmercaptoethanol with high [SDS] (1 or 2%) in the
sample buffer and try to heat the sample (with SB) 5 minutes at 100°C.
can U send me the results of these trials, for more explications
