Enough Methonine?

Hello all,

I am about to start an in vitro translation and would like to know two things:

1) I would prefer to use 35 S Methionine because it is readly available to me,
however my protein contains only one Methionine in its sequence. Is this enough
to be able to visulise the protein on a SDS gel?

2) For plant proteins is it best to use wheat germ extract to translate the
protein? I heard rabbit reticulocyte is sometimes better.

Thanks for your help in these matters,
Mike Mason
