protein purification lab

I am posting this for a friend who is teaching a biochemistry
metabolism lab this summer to fill in for the regular instructor who
has been in charge of the lab for many years (I'll be the T.A). The
protein purification lab is based on a paper from 1964 and involves
bucket load solvent extractions and other such fun steps. So we are
hoping to update this with another protocol that employs more
applicable techniques. Specifically, we would like the protocol to
include a gel filtration and an ion exchange chromatogaphy step, and
to be able to be completed over 3 four hour labs. Does anyone know of
any protocols which might be useful? We would really appreciate any
help, as time is short. We currently purify alkaline phosphatase, but
anything that has an easy-to-come-by source and an idiot proof assay
would be suitable.
