dear netters, I try to purify a solubilized his-tagged membrane protein. Binding of the protein on NTA is OK but it seems that only 10% of the protein can be eluted by a gradient of imidazole ranging from 30 mM to 0.5 M. I would like to know if other persons had the same problem. thank you
