I have been attempting to transfect primary endothelial cells
(HUVEC) by a variety of means but without much success.
Thus far I have used DEAE dextran, polybrene and lipofectamine
and although the procedures always work in COS cells results
are always negative in HUVECS. There are reports in the literature
using the above methods in HUVECS but others saying that the only
way to transfect HUVECS is by electroporation.
Anyone out there with any experience of tranfecting primary endothelial
cells ? Any protocols, suggestions or advice would be much appreciated.
