Just a note to tell that N-terminal blocking is indeed a very struggling
problem. The question is where you are interested in. Do you really want
the N-terminus, or just some sequence information to get DNA-probes ? If
the last is the case, you can always perform an 'in situ' digestion of the
piece of membrane and try to elute the obtained peptides, separate them by
HPLC and get sequences of them. there are many papers describing this
procedure.
If you are really interested in the N-terminus, try acylaminopeptidase; or
pyroglutamase, but be aware : they usually don't work.
