Hello!

I used extensively NTCB during the work for my Ph. D. thesis. Cleavage is 
reliable, but it's difficult to get a complete hydrolysis without having 
some unspecific cleavage. You can see the basic recipes in two of my 
papers, and their references:  Altamirano et a., (1992) Biochemistry 31 
1153-1158, and  (1993) Biochem. J. 295:645-648.  In the second paper, I 
used the reagents NSTB/sulfite to cleave and titrate disulfides 
(Thannhausser et al., reagent). I tried also NTCB/cyanide for the same 
purpose (cleavage and cyanylation of a S-S bridge). It works, but is not 
good to measure disulfides (NSTB/SO3= is better) because  the unstability 
of NTCB at pH higher than 8.
