What is the current opinion about which is the best E.coli genotype
for the production of missense mutant forms of eucaryotic proteins?
We are working with several derivatives of a eucaryotic protein
(the photoprotein aequorin), each of which is produced in E.coli
at levels significantly below that of a wild type form. Since I
assume the problem is due to protein degradation, we intend to
try using strains having other genotypes (eg. lon, htpR). If you have
experience with this type of problem, please send your comments.
I will summarize the responses and repost them if requested.
