we are getting some problems in genotyping by PE-ABI fluorescent
microsatellites and I will appreciate a reply from people using the same
set of markers.
1. most of markers need to be optimize for Mg concentration (i.e. not
working in the specified condition)
2. some markers show hill-shaped allele peaks, difficult to label in
heterozygotes.
3. allelic frequencies need to be calculated, referring to indicated CEPH id.
The enzyme used in PCR (Amplitaq or Taq Gold) seems not to change the
result quality. Suggestions are welcome.
