I would like to ask you two questions.
1. For the formation of its chromophore GFP needs oxygen. In theory this
would mean that GFP fluorescence can be used as an oxygen sensor. Does
anybody know or can indicate me a reference where to find how low the
oxygen pressure has to be before the chromophore is no longer formed? Do
you think that GFP fluorescence, i.e. presence or absence can be used as a
proof for presence or absence of oxygen?
2. I want to express GFP in insect cells from an intron containing gene to
be sure that there is no bacterial expression. Is a gene for GFP with an
intron which will be spliced out in insect cells available.
