GFP fusion proteins

Hi Dinakar:  In response to your question regarding GFP-fusion proteins in
Arabidopsis:  I have generated transgenic Arabidopsis expressing a
GFP-fusion protein using Agrobacterium mediated transformation (vacuum
infiltration method).  The plants appear healthy and fertile and the fusion
protein localizes to a subcellular compartment consistent with the result
obtained using a GUS-fusion protein.  The fusion was constructed using the
mGFP4 clone that you mentioned.  I adapted an expression plasmid containing
a CaMV 35S promoter to generate C-terminal fusions (mGFP-X), and didn't use
the Clontech vectors.  Trying to express the unmodified GFP cDNA (rather
than mGFP4) in Arabidopsis using particle bombardment has met with no
success, although both the unmodified GFP and GFP-fusions can be expressed
sucessfully in other plant species.
