Fluorescent Staining Protocol...

     I was hoping someone could provide me with a reference for a 
differential fluorescent staining protocol to differentially stain RNA 
from DNA located in the polytene chromosomes.  I am working with a fellow 
diptera Chironomus riparius, and am looking at puff induction.  
Previously, I have tried DAPI (not differential but fluorescent) and 
acridine orange (Nash and Plaut, 1964), but neither was to my 
satisfaction.  Recently I have come across a reference using 
fast-green/orcein, but I haven't research its use as of yet.  The stain 
is to be used on the salivary gland tissue of fixed (most likely Carnoy 
solution, possible something else) chironomids.
