I have a question about counterstaining polytene chromosomes following in situ
hybridization with a biotinylated probe. I am using the ENZO Detek Hrp system
for color generation, and when I counterstain with Giemsa the banding pattern
is not as sharp as I would like. The protocol that comes with the kit
suggests to counterstain with fast green, but they don't say for how long or
at what concentration. if anyone could help me I would greatly appriciate it.

thanks in advance
