Labelling with amino acids

Does anyone have experience labelling Chlamydomonas in vivo with
radiolabelled amino acids (i.e. 3H or 14C)?  Will the amino acids be taken
up from the culture media in sufficient quantity to do a pulse labelling of
protein synthesis?  Is it necessary to toluene-permeabilize the cells to
accomplish this?  Any protocols, tips, hints, etc. will be greatly
appreciated.  Thanks in advance.
