Hi, I am encountering a problem trying to deduce the 5' end sequence
of a parainfluenze virus. I have tried the Clonetech kit and didn't
obtained a good result. Then I checked the published sequence of PIV
and found that there might be some secondary sturctures that could
interfere with my RT reaction. I have also tried the T4 RNA ligase
method with a short piece of DNA oligo (I am still waiting on my RNA
oligo) but it didn't work possibly due to the low efficiency of the
RNA ligase working with a DNA oligo.
So my question is that has anybody out there who can give me some
pointer on how they did it??? Or some information on where I could
find some good literature??? Thanks
