You don't *have* to use carbonate buffer for coating
ELISA plates (I usually use PBS for diluting my samples).
The optimum buffer will probably depend on your antigen
and on the type of plate that you are coating. It sounds
like your purified fractions should be fine but you may
want to do a quick test with some authentic protein.
