Unit of enzyme...

Dear everyone:

If we buy a enzyme, we can see the label which say how the producing
company determines the unit of it; the substrates, the buffer condition,
the temperature, etc.. But except special case, for example, restriction
enzyme(it is always packed with the buffer for it), we usually make
buffer different from the buffer, which company used, for the enzyme
reaction. In this point, do we recheck and adjust the unit per mg for
our own reaction buffer? That is, the unit which is described in papers
is the unit that was re-check by their own buffer system, not written
on the reagent's label?

I did not considered because I did not work with enzyme until now.
