An old protocol utilized multiple freeze-thaw cycles on a plasmid-agro
suspension (I think cells were made comp with CaCl2). Never tried it
myself, but a very experienced colleague regularly used to he says, and
does sometimes now. The lab is pretty much settled on triparentals these days.
