a postdoctoral position is available starting July 1st 1994 for 2 1/2 years
(until 31/12/96) to establish an Arabidopsis BAC library and to create a
physical map of overlapping clones. The BAC (bacterial artificial chromosome)
cloning system is based on a F-derived vector that allows to clone and stably
propagate fragments of DNA up to 350 kb in E.coli (see Shizuya et al.,
PNAS 89, 8794-8797, 1992). Due to their easy handling (similar to cosmids),
their high stability and the low fraction of chimeric clones, BACs will be a
highly useful tool for a variety of applications including positional cloning
of genes / complementation of mutants and genome sequencing.
Applicants should have expertise in molecular cloning techniques especially
in cloning and handling of large (plant, animal, or human) DNA fragments
(YACs, BACs, P1).
