I checked: If the rat strain used is similarto the one for which data
of  ploidy as a
function of age exists, then 5 months is just about enough time to get
significant tetraploids (which have double the volume as mononucleate
diploids). But the numbers are not consistent with any zonation of
ploidy, so I may be misinformed on this issue.
